Commander Shepard saw Garrus at the Citadel Tower, arguing with Executor Pallin and asking for more time to finish his investigation. Garrus told Shepard that because of Saren's Spectre status it was nearly impossible for him to get anything useful, but he knew in his gut that Saren was guilty. Though Garrus was told his investigation was over, he decided to defy the executor's order and pursue another lead on his own.

Otherwise Shepard meets him in the C-Sec offices on the Citadel, baiting one of the officers.
Wrex initially joins Shepard because he has been hired to kill Fist by the Shadow Broker. After Fist is taken care of, Wrex remains with Shepard either out of gratitude, or because he wants to be in the thick of the action as Shepard seeks out Saren Arterius. As Wrex puts it, "a storm's coming, and you and Saren are right in the middle of it."
Joker
Details:
The sarcastic but brilliant pilot Joker was helmsman of the Normandy SR-1 and helped Shepard defeat Saren and save the Citadel. Cocky because he knows his own talent, Joker is one of the best pilots in the galaxy.
Part of Joker's outlook on life is shaped by an ailment he has suffered his whole life, brittle bone disease. While able to walk and function relatively normally, small injuries could be fatal to him. The recoil from a rifle can cause fractures, and a fall can easily break bones. Piloting a ship gives him a freedom he can't get anywhere else, as he is too fragile to pursue physical endeavors others take for granted. He lives to fly, and rarely leaves the ship.
David Anderson
Details:
David Anderson was once captain of the Normandy SR-1. Once Commander Shepard became the first human Spectre, Anderson respectfully handed over the ship to him to track down Saren and his Geth army. Anderson is one of the Alliance's most decorated special forces operatives, and one of the first to graduate from the N7 marine program. Additionally he served with honors during the First Contact War between the turians and the Alliance. Because of his impressive service record he was the first human to be considered for the Spectre program. When a mission with a fellow Spectre, Saren, went terribly wrong, he was blamed and the fiasco was covered up.
Though he loathes dealing with bureaucracy in all its forms, Anderson now resides on the Citadel playing a role in galactic politics.
The Illusive Man
Details:
Mysterious and powerful, the Illusive Man commands Cerberus, a secretive pro-human organization dedicated to protecting mankind's interests in the galaxy. He believes the ends justify the means, at any cost. Nothing is more important than ensuring not only humanity's survival in the galaxy, but also its dominance. To combat the new Collector threat, the Illusive Man recruits Commander Shepard and gives him the tools to build the team needed to save mankind. Is there something behind his altruistic façade, and more importantly can Shepard trust the Illusive Man?
